Research Focus

Although optical microscopy has evolved over centuries, it grapples with dual challenges: phototoxicity conflicting with the demand for extended and high-speed observations, alongside the contradiction between the non-uniform refractive indices and the requirement for high spatiotemporal resolution. I am committed to the interdisciplinary research with a specific focus on pioneering breakthroughs in computational imaging and intelligent microscopy. In terms of hardware, I enabled comprehensive recordings of temporal, spatial, and angular information for high-speed, high-resolution 3D microscopy including scanning light field imaging, confocal scanning light field imaging, achieving the milestone of continuous three-dimensional observation across 300,000 consecutive time frames in mammals. As a result, the international record for continuous observation was extended from minutes to an impressive 48 hours. In terms of algorithms, I developed artificial intelligence technologies to enhance the speed, resolution, and robustness of fluorescence images and videos, with contributions including super-resolution, 3D reconstruction, denoising, de-scattering, aberration removal, and motion correction through phase-space correlation modeling, achieving a 20 dB improvement in signal-to-background ratio, a 700-fold increase in computation speed, and a 120-fold reduction in memory consumption, which makes large-scale, high-fidelity data processing practical. These breakthroughs solve the long-standing challenge of dynamically tracking the high-speed behavior of immune cells in complex intravital environments, pioneering whole-brain functional studies based on membrane potential signals at single-neuron resolution. My future research will further focus on mesoscopic imaging and analysis, computational neuroscience, multimodal understanding and intelligent analysis, and AI for science.
